Glycosaminoglycan Changes in Regenerated Menisci of the Immature Rabbits -Histochemical and Spectrophotometric Quantitation-


Abstract
Meniscus in the knee joint has optimal structure for lubrication, load transmission, stabilization of joint, distribution of external force and protection of articular cartilage. After total meniscectomy, it was reported that degenerative changes
develop due to its absence in case of non-forming which results in decreased contact area between femoral and tibial articular surface. Even in the case of the meniscal regeneration, the regenerated meniscus cannot perform full function because
of
its
inadequate morphology, histology and chemistry. Glycosaminoglycan in the ground substance of meniscus provides high elasticity and ability to endure compressive and distractive stresses. However, the role of the glycosaminoglycan in the meniscus
has not
been well documented.
Therefore this study was conducted primarily to compared the contents and distribution of glycosaminoglycan in the normal and regenerated meniscus, and partly to elucidate the role of glycosaminoglycans in the meniscus to fulfil its function.
Sixty immature rabbits, weighing 300-400 gm and aged 4 weeks, were used for the study. Total medial meniscectomy in the right knee joint was performed in all rabbits. The medial menisci of left knee joints were used as control. After 16, 20, 24,

weeks postoperatively, the amount of glycosaminoglycan in normal and regenerated menisci were measured. For the measurement of quantity and distribution of glycosaminoglycan contents, photoabsorptiometry and safranin 0 staining method were used.
@ES The results were as follows:
@EN 1. In fourty-eight of the sixty medial meniscectomized knees, menisci were regenerated in various sizes with degenerative changes of joints, especially on tibial side.
2. In normal menisci, glycosaminoglycan was distributed at inner and middle 1/3, while glycosaminoglycan in regenerated menisci was diffusely found in entire width.
3. The concentrations of glycosaminoglycan per 1 mg of regenerated menisci were 4.36¡¾1.68¥ìg, 4.58¡¾1.14¥ìg , 4.65¡¾2.21¥ìg , 4.60¡¾2.06¥ìg in each 16, 20, 24, 28 weeks groups, while those of normal menisci were 9.10¡¾2.09¥ìg, 9.75¡¾3.97¥ìg ,
11.80¡¾8.44¥ìg , 14.51¡¾8.91¥ìg.
4. In normal menisci, the amount of glycosaminoglycan was gradually increased as rabbits getolder. However, there was no statistical differences of quantitative amount of glycosaminoglycan of regenerated menisci amoung the 16, 20, 24, 28 weeks
groups
during the observation periods(P>0.05).
5. There was a distinctive difference in safranin 0 stainability by every 4¥ìg increase of glycosaminoglycan in both 1 mg of normal and regenerated menisci.
This study showed the quantitative change of glycosaminoglycan between normal and regenerated menisci, and comparative analysis of glycosaminoglycan contents in the menisci by biochemical method and histological method were found to be useful
tool.
